چکیده

Abstract In preclinical studies, simple coumarins (scoparone, limettin) and furanocoumarins (imperatorin, xanthotoxin, bergapten) have already found to demonstrate procognitive abilities. This suggests that they hold antioxidative, anti-inflammatory inhibitory action towards acetylcholinesterase activities. However, little is known about their influence on the amyloidal structure formation, leading cause of Alzheimer’s disease (AD). vitro in cellulo assays were applied evaluate effect selected different stages Aβ40/42 tau protein aggregation. Kinetic analyses performed inhibiting abilities time. Limettin revealed most potent profile Aβ40 aggregation, however, all tested compounds presented low Aβ42 aggregation inhibition. Despite preliminary stage project, promising effects shown. coumarin scaffold can serve as a potential multitarget agent AD treatment, but further studies are required confirm this.

MOAB-2 Mouse Monoclonal antibody to Amyloid beta peptide (A beta 40/42), purified

Catalogue No.: M-1586-100 Description: The amyloid beta peptide is derived from the cleavage of the Amyloid precursor protein (APP) and varies in length from 39 to 43 amino acids. However, the form(s) of amyloid-beta peptide (Aß) associated with the pathology characteristic of Alzheimer’s disease (AD) remains unclear. In particular, the neurotoxicity of intraneuronal Aß accumulation is an area ...

Amyloid beta -protein (Abeta) assembly: Abeta 40 and Abeta 42 oligomerize through distinct pathways.

Amyloid beta-protein (Abeta) is linked to neuronal injury and death in Alzheimer's disease (AD). Of particular relevance for elucidating the role of Abeta in AD is new evidence that oligomeric forms of Abeta are potent neurotoxins that play a major role in neurodegeneration and the strong association of the 42-residue form of Abeta, Abeta42, with the disease. Detailed knowledge of the structure...

In vitro and in vivo protein oxidation induced by Alzheimer's disease amyloid beta-peptide (1-42).

Amyloid β-peptide (Aβ) is thought by many researchers to be central to the pathogenesis of Alzheimer’s disease (AD) (reviewed in Ref. 1). In addition, oxidative stress, manifested by protein oxidation and lipid peroxidation, is apparent in AD brain.2,3 Our laboratory developed a comprehensive hypothesis for neurotoxicity in AD brain that unites these two observations and provides a testable fra...

Shuttling protein nucleolin is a microglia receptor for amyloid beta peptide 1-42.

Amyloid-beta peptide 1-42 (Aβ42) plays a key role in the neurotoxicity found in Alzheimer's disease. Mononuclear phagocytes in the brain (microglia), can potentially clear Aβ via phagocytosis. Recently, the shuttling-protein nucleolin has been shown to possess scavenger receptor-activity. Here, we investigated whether this receptor interacts specifically with Aβ type 1-42 and mediates its phago...

Amyloid beta 42 peptide (Abeta42)-lowering compounds directly bind to Abeta and interfere with amyloid precursor protein (APP) transmembrane dimerization.

Following ectodomain shedding by beta-secretase, successive proteolytic cleavages within the transmembrane sequence (TMS) of the amyloid precursor protein (APP) catalyzed by gamma-secretase result in the release of amyloid-beta (Abeta) peptides of variable length. Abeta peptides with 42 amino acids appear to be the key pathogenic species in Alzheimer's disease, as they are believed to initiate ...
